Destounis made his debut in 1964 with the "Arma Theatrou" troupe and went on to collaborate with various theatrical troupes, including those of Dimitris Myrat, Mimis Fotopoulos, and Sofia Vembo. He began his career in cinema with the film A Ship Papadopoulos in 1966 and has also appeared in television plays and series.
